    Speaker 梁文轩,中国科学技术大学生物医学工程学院与物理学院双聘特任研究员,博士生导师, 入选中科院青年人才项目。本硕毕业于清华大学生物医学工程系,博士毕业于约翰斯霍普金斯大学生物医学工程系,入职中科大前在哥伦比亚大学祖克曼研究所(Zuckerman Institute)开展博士后研究。长期致力于面向活体组织的先进光学显微成像方法与仪器研究,在Light: Science & Applications、IEEE Transactions on Medical Imaging、Nature Biomedical Engineering、ACS Photonics 等期刊上发表论文20余篇(其中一作/共一/通信8篇),谷歌学术h指数为16,并有3项发明专利已获授权。 Time 2023/08/16 10AM Location Room 1-418, SEIEE Building Host Yuye Ling, Associate Professor, Department of Electronic Engineering Abstract 活体三维荧光显微成像旨在实时、原位、无创或微创地观测活体组织及模式动物体内的三维(亚)细胞结构并跟踪其功能动态,在生命科学(尤其是神经科学)和临床诊疗两方面都大有可为。相比于离体组织切片或培养细胞,活体组织为成像技术带来了新的挑战:一是需要具备光学层析能力以直接探查厚样本,二是具备足够的时空分辨率以有效对抗活体组织运动的干扰,三是需要尽可能高的探测灵敏度以最小化光剂量与光毒性,四是需要发展小型化、微型化的成像设备以实现内部器官与组织的在体原位探查。围绕以上挑战,报告人开展了系列研究:1)研制了面向临床实时在体荧光成像的微型双光子显微内窥镜和小型化共焦斜光片显微镜(SCAPE),通过架构设计与物理原理层面的创新,取得了媲美标准台式设备的成像性能;2)研发了面向模式动物全脑神经成像的大视场SCAPE显微镜,提出了基于纤维光锥的跨介质中间图像耦联机制,使得大视场SCAPE系统的荧光收集效率有数量级的提升,能够以约10体积/秒的时间分辨率和微米级别的空间分辨率对约4×4×0.5毫米长宽深的三维视场实施高速体积成像。
